C13DEPT Experiment

This experiment WILL interleave the normal 1D 13C
spectrum and the DEPT spectrum (i.e., acquire bs
transients of the first spectrum, then acquire bs
transients of the second spectrum, then repeat until all nt
transients have been collected). However, this requires that both spectra have
the same number of transients. If you prefer to collect a different number of
transients for each spectrum, then you can acquire a normal 1D 13C
spectrum and a DEPT spectrum in two separate experiments; however, if you use
two different experiments, you CAN NOT interleave the experiments.
- Type c13dept to set up the c13dept experiment.
- Select:
- "BB" (13C 1D) experiment.
- "BB/DEPT135" (13C 1D and DEPT 'CH-up,CH2-down, CH3-up') experiment.
- "BB/FULL DEPT" experiment.
- "DEPT ONLY" experiment.
- Enter the text that you want to print at the top of the printout.
- Verify the following parameters (listed at the console):
- pw ....... 90° pulse for 13C.
- pp ....... 90° pulse for 1H pp pulse.
- For Intermediate Users: verify that the following parameters are set correctly:
- j .......... average 1H - 13C coupling constant. The default is 140 Hz, or 125 Hz for aliphatics only, or 165 Hz for aromatics.
- d1 ....... relaxation delay for the standard 13C spectrum. d1+at should be (1~3)T1 of 13C. The default is 0.6 sec but if you want to see quaternary carbons in the 13C 1D spectrum, you may want to try setting d1 equal to 3.0 seconds.
- d2 ....... relaxation delay for DEPT. d2+at should be (1~3)T1 of 1H. The default is 0.6 sec.
- nt ........ number of transients. Multiples of 4 are needed with 16 being the best, 256 is the default setting.
- bs ....... block size or number of transients collected before switching to the next spectrum.
- Type au (or ga)
to acquire the spectra. The normal 1D 13C spectrum (or DEPT45
spectrum, depending on the experiment selection at step 2) will be displayed at every bs transients.
- OPTIONAL: You can stop the acquisition (e.g., type sa) before all nt transients have been collected.
- Type wft and phase your 13C 1D spectrum. The DEPT spectrum will use the same phasing as the 13C 1D spectrum.
- Type c13deptproc to automatically process and plot the spectra on one page. The spectrometer will ask if you want to print on 11x17 paper. Type y to select 11x17 paper, type n to use the device that the plotter parameter is set to.
- Save your experiment. You can save both spectra in one experiment with one file name. Pick up your printout.
